Murciano began his career in 1997 when he featured in the action film Speed 2: Cruise Control. In 2000 he appeared in the crime drama Traffic where he played the role of a DEA agent. He also starred in the soap opera Spyder Games in 2001 as Francesco Torres. In the same year, he appeared in the war thriller Black Hawk Down where he met Jerry Bruckheimer. Through him, he was offered a starring role as Danny Taylor in the CBS mystery drama Without a Trace. The role led to him being nominated in 2008 and in 2009 for the ALMA Award.
Murciano has also made appearances in the comedy Miss Congeniality 2: Armed and Fabulous, the Peruvian-Spanish drama Máncora, and How to Go Out on a Date in Queens. He also appeared in Yes We Can, a music video by Will. i.am. Murciano appeared in season 10 episode 4 called Coup de Gráce of the drama CSI: Crime Scene Investigation as Det. Carlos Moreno. He had a recurring role in April 2011 in the drama NCIS as CIA Agent Ray (CI-Ray) Cruz in the eighth season episode Two-Faced and episode Pyramid. Murciano had his last appearance in the series, in January 2012, in season 9 episode 13 called A Desperate Man.
He returned to CSI: Crime Scene Investigation in April 2012 reprising his character as Det. Carlos Moreno in the 12th season in the episode Altered Stakes. Murciano had a recurring role in the horror drama 666 Park Avenue playing Dr. Scott Evans. He also co-starred in the film Dawn of the Planet of the Apes the sequel in 2014, as Kemp. He played the role of Felipe Lobos in Power from 2014 to 2016. Murciano became a series regular in 2015 when he played Det. Marco Diaz on the series Bloodline. In 2007, he appeared in the Netflix film Bright playing the role of a gang leader called Poison.

When he was a teen he became a drug addict and alcoholic as a result of the incident.
