Later, as he was was still working in Neighbors, he co-starred in several films like Uptown Girls, Winning London, and Swimming Upstream. Spencer appeared in the FOX medical dram House, where he starred as surgeon and intensive care specialist Dr. Robert Chase from 2004 to 2012. He played the character for the whole run of the show, and this made him the 2nd longest serving member of the team.
In 2012, Spencer starred in the NBC drama Chicago Fire playing the role of Matthew Casey, till 2021. It was announced in 2022, that he would return to the television series Neighbors, in the series finale. He began his career in music with the Australian Boys Choir in which they performed together from the year 1986 to 1992. Spencer plays the violin in a group where Hugh Laurie his former co-star from House is a member, called Band from TV. The group plays at events where they earn money and donate it to charity. In April 2008, the group played on Idol Gives Back.
Spencer was a nominee for Most Popular Actor at the Logie Awards in 1998 and 1999, due to his role in Neighbors. He was also nominated for Choice TV Breakout Performance – Male at the 2005 Teen Choice Awards, for his role in House.
