Squerciati was awarded the Agnes Moorehead award for her performance in the play Just in Time: The Judy Holliday Story as Judy Holliday, at the  Lucille Lortel Theatre. She Broadway debut was in To Be or Not to Be, a Ernst Lubitsch’s adaptation that was directed by Casey Nicholaw, a Tony Award winner. Squerciati starred in the off-Broadway, Manipulation, Beauty of the Father among others. She created a role in the cappella musical comedy Perfect Harmony, called Kerri Taylor, and she was also a member of The Essentials.

Squerciati played the role Polichinelle in the film The Nutcracker, which was directed by Emile Ardolino in 1993. Her feature film debut was in It’s Complicated, which she did in the year 2009. She later appeared in the films  Frances Ha and Alter Egos  in 2012, and also starred as Dawn in the indie film Sparks which is based on the comic SPARKS by William Katt. Her first role on television was Betsy Naylor on Law & Order: Criminal Intent in the year 2009. In 2010 and 2011, Squerciati appeared in guest roles on Damages, The Good Wife, Law & Order: Special Victims Unit, and Blue Bloods. She gained popularity in season five of Gossip Girl that has eight episodes. She then guest starred as Soviet spy in the series The Americans.

It was announced in August 2013 that Squerciati would appear in the television series, Chicagi Fire spin-off Chicago P.D. as Officer Kim Burgess. The series debuted on 8th January 2014 and its seventh season premiered on 25th September 2019. NBC later announced on 27th February 2020 that the series was being renewed for season eight, nine, and 10. She also appeared in the television series Special Skills in 2017, taking the role Marina.
